Изучить распределение аллелей и генотипов полиморфизма I/D по гену ангиотензин-превращающего фермента (АСЕ), полиморфизма M235Т по гену ангиотензиногена (AGT), полиморфизма А1166С по гену рецептора ангиотензина II 1-го типа (AGTR1) в группе детей с артериальной гипертензией (АГ) и в группах сравнения. Материалы и методы. Обследовано 84 пациента с АГ, 146 детей с нормальными значениями артериального давления (АД) и 398 детей, представляющих случайную выборку. Результаты. Выявлено статистически значимое повышение частоты встречаемости аллеля 235Т в группе детей с АГ по сравнению с детьми из группы с нормальными значениями АД. Обнаружено повышение частоты генотипа ТТ в группе детей с АГ по сравнению с распространенностью этого генотипа у детей двух групп сравнения. У девочек с АГ обнаружено достоверное повышение частоты аллеля 235Т и генотипа ТТ гена AGT по сравнению с их частотами у девочек из групп сравнения. Среди мальчиков с АГ выявлено значимое преобладание генотипа ТТ и аллеля 235Т гена AGT по сравнению с мальчиками с нормальными значениями АД. Достоверных различий в распределении аллелей и генотипов полиморфизма I/D гена АСЕ и А1166С гена AGTR1 у детей исследуемых групп не выявлено. Заключение. Необходимы дальнейшие исследования с увеличением объема выборки обследованных детей с АГ, подтвержденной результатами суточного мониторирования АД, для уточнения роли полиморфизма М235Т полиморфизма гена ангиотензиногена в развитии АГ у детей.</p></abstract><trans-abstract xml:lang="en"><p>Objective. To study the I/D polymorphism of angiotensin converting enzyme gene (ACE), the M235T polymorphism of the angiotensinogen gene (AGT) and the A1166C polymorphism of the angiotensin II type 1 receptor gene (AGTR1) in children with arterial hypertension (AH) and in controls. Design and methods. 84 children with the diagnosis of AH, 146 children with normal values of blood pressure and 198 children of population sample were included in the study. АН was defined as systolic/diastolic blood pressure measurements higher than 95 age-gender-height percentile of the adopted reference values. DNA was extracted from blood samples according to standard protocols and analyzed by the PLFP method. Results. The frequency of T235 allele of the AGT gene was significantly higher in hypertensive patients than in children with normal values of blood pressure. The frequency of TT genotype in hypertensive patients was significantly higher than in two control groups. The frequency of T235 allele and TT genotype were significantly higher in group of hypertensive girls compared with girls of two control groups and in hypertensive boys compared with normotensive boys. The distribution of ACE genotype and allele frequency did not differ significantly between patients with АН and control subjects. There were no significant differences in the frequency of CC genotype and C allele of AGTR1 gene in children with АН compared with control subjects. Conclusion.
