В основе патогенеза сердечно-сосудистых заболеваний лежит хроническое системное воспаление. Точные механизмы, вызывающие развитие гипертензии, неясны, но есть сведения об участии в нем медиаторов воспаления. Целью настоящего исследования было изучение роли межгенных взаимодействий полиморфных маркеров генов IL6, IL1B, IL12B, TNF, LTA и IL10 в развитии предрасположенности к эссенциальной гипертензии (ЭГ). Результаты. В этнической группе татар, проживающих в Республике Башкортостан, нами обнаружена ассоциация с заболеванием полиморфных маркеров rs16944 гена IL1B и rs1800872 гена IL10. В результате анализа ген-генных взаимодействий выявлено наличие значительного синергизма между полиморфными маркерами rs3212227 гена IL12B и rs1800629 гена TNF в отношении ЭГ. В итоговую модель, построенную с использованием логистического регрессионного анализа, в качестве оптимальных предикторов ЭГ вошли полиморфизм rs1800872 гена IL10 и сочетания полиморфных маркеров IL12B rs3212227 и гена TNF rs1800629. Заключение. Полученные нами данные позволяют сделать вывод о вовлеченности цитокиновой системы в патогенез ЭГ. </p></abstract><trans-abstract xml:lang="en"><p>Background. Chronic systemic inflammation is a key factor in the pathogenesis of cardiovascular diseases. The exact mechanisms underlying blood pressure elevation are unclear, but recent findings suggest that inflammatory mediators are involved. The objective of our study was to investigate the influence of gene-gene interactions between IL6, IL1B, IL12B, TNF, LTA and IL10 genes polymorphic markers on susceptibility to essential hypertension. Results. In the ethnic group of Tatars residing in Bashkortostan, Russia, we found an association of IL1B rs16944 and IL10 rs1800872 polymorphic markers with the disease. The analysis of gene-gene interactions showed significant synergy between IL12B rs3212227 and TNF rs1800629 polymorphic markers towards essential hypertension. The final model (logistic regression) included IL10 rs1800872 genetic marker and the combination of IL12B rs3212227 and TNF rs1800629 genetic markers as optimal predictors of essential hypertension. Conclusion.
